ABSTRACT

Objective:To explore the relationship between drug evidence and core prescription for depression.Methods:We retrieved literature of TCM for depression from CNKI, VIP and Wangfang databases to November 2019, 30th as well as there cords from Ancient and Modern Medical Records Cloud Platform (V 1.5). The Excel 2010 was used to establish the standardized database of medical records. After the standardization of medicines, Ancient and Modern Medical Records Cloud Platform (V1.5) statistics methodswere used for association rules analysis, complex networks, and analysis of drugs’ frequency, medical characteristics, core prescription drugs.Results:A total of 632 effective prescriptions were included, involving a total of 527 drugs. The results of frequency of herbs showed that 23 kinds of high-frequency herbs were obtained. Bupleuri Radix was the most frequently used medicine. Most herbs are warm or flat, with pungent, sweet and bitter in taste, belonging to the lung, liver, heart and spleen meridians. A total of 25 drug-pair association and 13 TCM association were obtained by association rule analysis. Conclusions:TCM treatment for depression is mainly based on soothing the liver and regulating qi, clearing the heart and calming the nerves. Bupleuri Radix, Curcumae Radix, Paeoniae Radix Alba, Chuanxiong Rhizoma, Ziziphi Spinosae Semen are the basic prescriptions. Angelicae Sinensis Radix, Ophiopogonis Radix, Albiziae Cortex, Polygalae Radix, Poria are used as reference.

ABSTRACT

ObjectiveTo investigate the mechanism of action of Sini powder in the treatment of liver cancer based on network pharmacology and molecular docking. MethodsTraditional Chinese Medicine Systems Pharmacology Database and Analysis Platform was used to obtain the compound and target of Sini powder, and the corresponding gene Symbol was obtained through Uniprot. The disease genes of liver cancer were obtained from Human Genome Database, and the genes with intersection with the target genes of Sini powder were screened out. Cytoscape3.7.1 software was used to draw the map of “traditional Chinese medicine (TCM)-compound-target” network. STRING was used to construct a protein-protein interaction (PPI) network, R studio software was used to conduct gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses on therapeutic targets, and then the results were visualized. The active component with the highest number of targets was selected as the ligand, and the target with the highest degree in the PPI network was selected as the receptor, so as to predict the structure of receptor-ligand complex and the amino acid residues that bind to each other. ResultsIn this study, 91 core targets and 141 relevant active components of Sini powder were screened out, among which quercetin and kaempferol were the main active components in the treatment of liver cancer. TP53 and HSP90AA1 were the main therapeutic targets. The GO enrichment analysis obtained 1007 items which met the screening criteria, which were mainly involved in the biological processes of antioxidation reaction, activity regulation of protein serine and threonine kinase, and cellular stress response. The KEGG enrichment analysis obtained 102 pathways, which mainly regulated the hepatitis B pathway and the PI3K-Akt signaling pathway in the prevention and treatment of liver cancer. The results of molecular docking showed a synergistic antitumor effect between the crystal structure domains VAL147, CYS220, GLU221, and PRO222 of quercetin-TP53. ConclusionThis study reveals the mechanism of action of Sini powder in the treatment of liver cancer by acting on multiple targets and signaling pathways, which provides a theoretical basis for biological experiments.

ABSTRACT

Objective:s To evaluate the impacts of prior surgical scores(PSS) on the clinical efficacy and perioperative safety of cytoreductive surgery(CRS) and hyperthermic intraperitoneal chemotherapy(HIPEC) for pseudomyxoma peritonei(PMP).Methods:From the comprehensive PMP database, we collect the cases treated for the first time by CRS+ HIPEC, to form this study cohort. The clinicopathological features, PSS, CRS+ HIPEC details, overall survival(OS), and serious adverse events(SAEs) are systematically analyzed, to study the correlations between PSS and OS or SAEs.Results:335 PMP cases received standardized CRS+ HIPEC in this study. The median OS is 58.2 months for PSS-0 patients, 63.7 months for PSS-1, and 55.4 months for PSS-2/3, with no statistically significant differences in OS among the different PSS groups(χ 2=0.499, P=0.779). Subgroup analysis by pathologic types also found no statistically significant differences among the different PSS groups. Moreover, no significantly statistical differences are observed in overall SAEs(χ 2=0.625, P=0.722), CRS-related SAEs(χ 2=0.267, P=0.901), and non-CRS-related SAEs(χ 2=0.677, P=0.715), among the different PSS groups. Conclusions:PSS does not pose significant impacts on the efficacy and safety of CRS+ HIPEC for PMP patients at experienced treatment center.

ABSTRACT

Objective@#To establish the patient derived xenograft (PDX) model of pseudomyxoma peritonei (PMP), and identify the key characteristics of tumor biology of this model, in order to provide a reliable model for studying the pathological mechanisms and new therapeutic strategies of PMP.@*Methods@#PMP tumor tissue was obtained from surgery and cut into pieces after washing. Then tumor pieces were implanted subcutaneously in BAL B/c-nu mice for 6 stable passages. In the 7th passage, tumor tissue was implanted orthotopically into abdomen. Subcutaneous tumor and orthotopic tumor were then homogenized to make tumor cell suspension, implanted into abdomen of 10 BAL B/c-nu mice through midline laparotomy, 100 μl for each. The key experimental parameters including body weight changes in the observation period, experimental peritoneal cancer index (ePCI) score at the autopsy, histopathological and immunohistochemical characteristics, and gene expression profiles by high-throughput whole-genome exon sequencing were detected and recorded.@*Results@#The successful rate of established orthotopic PDX model of human PMP was 100% (10/10). The animals showed smooth body weight increases after tumor inoculation until day 27, then the body weight began to decrease steadily. Widespread tumor dissemination of PMP tumor through the whole abdomen was found by autopsy, including the diaphragm, liver, spleen, stomach, kidney, parietal peritoneum, bowel and mesenterium. Gelatinous ascites was also observed in abdominopelvic cavity. The ePCI score ranged from 5 to 9, with a 8 of median ePCI. Histopathological studies showed peritoneal mucinous carcinomatosis accompanied with signet ring cells (PMCA-S), obvious tumor cell atypia and parenchymal invasion.Immunohistochemistry showed the expressions of MUC1, MUC2, MUC5AC, CEA, CA199, CK20, CDX-2 and Ki-67 were positive, MUC6, CK7 and p53 were negative. Whole-exome sequencing identified that the most significant genetic alteration is the exon10 missense mutation c. 1621A>C of KIT gene, the mutation abundance was 89.7%.@*Conclusion@#PDX model of PMCA-S is successfully established, which displays the characters of high-degree malignancy, high proliferation and strong aggressiveness.

ABSTRACT

Objective@#To analyze the efficacy and safety of cytoreduction surgery (CRS) combined with hyperthermic intraperitoneal chemotherapy (HIPEC) for pseudomyxoma peritonei (PMP) in the early stage.@*Methods@#The clinical data, including pathological features, recurrence and survival of 65 PMP patients in the early stage underwent CRS combined with HIPEC in Aerospace Center Hospital from January, 2011 to December, 2018 were retrospectively analyzed.@*Results@#65 patients with early stage PMP underwent CRS+ HIPEC. Among these patients, 25 were males and 40 were females, and the mean age was 52.5 years. The median peritoneal cancer index was 3 (0-16). The score of completeness of cytoreduction (CC) of 63 patients (96.9%) was 0, and 2 patients (3.1%) was 1. No perioperative death occurred, the incidence of surgical complications above grade 3 was 3.1%. Three patients relapsed during the follow-up period, including 1 patient with low-grade PMP, 1 patient with high-grade PMP, and 1 patient with high-grade PMP accompanied by signet ring cell. The 5-year disease-free survival rate of the whole group was 92.4%.@*Conclusions@#PMP patients in the early stage treated by CRS combined with HIPEC can achieve benefit and safety. A close long-term follow-up is necessary.

ABSTRACT

Objective: To investigate the clinicopathological features and treatment strategy of pseudomyxoma peritonei (PMP) of ex-tra-appendiceal origin. Methods: Clinical data of 34 patients diagnosed with PMP of extra-appendiceal origin who were treated by cy-toreduction surgery (CRS) and hyperthermic intraperitoneal chemotherapy (HIPEC) in the Aerospace Center Hospital from September 2011 to February 2019 were retrospectively analyzed. Clinical and imaging features were summarized and the Log-rank test was used for survival analysis. Results: The clinical manifestations of the 34 patients with PMP of extra-appendiceal origin were mainly abdomi-nal distension (58.8%) and abdominal pelvic mass (52.9%), which are very similar to those of appendiceal PMP. The incidence of main complications after CRS and HIPEC was 14.7%. During the follow-up period of a median of 12 months (range 1-46 months), 9 patients died, and the 1-and 3-year overall survival rates were 69.6% and 53.5%, respectively. In the univariate analysis, peritoneal cancer in-dex (PCI)>20, no HIPEC, and non-radical surgery were significant risk factors for poor prognosis, while gender, age, origin, and patho-logical type did not show significant correlations. Conclusions: The clinical features of PMP of extra-appendiceal origin are not differ-ent to those of PMP originating from the appendix. It is difficult to ascertain the primary lesion before the operation; however, regard-less of the origin, CRS combined with HIPEC is always a safe and effective treatment choice.

ABSTRACT

Extra-articular distal tibial fractures as a result of high-energy damage are often comminuted or displaced, frequently accompanied by severe soft tissue injuries.Poor blood supply and various complications make the treatment more difficult,affecting life quality of the patients.The main goals of the treatment are to abtain a healed,well-aligned fracture,functional range of motion of the ankle joint and minimizing complications.It is generally recommended that surgical treatment be performed in the proper context of local conditions to facilitate early functional exercise.Plate fixation and intramedullary nail fixation are the common options for closed fractures.This article focuses on the two treatment methods and some important auxiliary technologies in both domestic and foreign, hoping to provide some references for clinical treatment.

Objective To analyze the clinical effect of a bifurcated ball frame designed for undergraduate research to assist hand-knee position,avoid physical exertion and increase comfort.Methods Randomized controlled trials consisted of 260 occipital (posterior) primiparous women born during labor,and were randomly divided into the observation group and the control group with 130 cases each.The observing group's mothers were on the yoga ball on the device.The angle and height of the frame and the ball plate were adjusted according to the maternal body type,so that the yoga ball at the chest and the cross could effectively support the maternal weight loss knee burden,and both sides of the small abdominal yoga ball could be effectively posted.After the adjustment was completed,the maternal relaxes and gently shakes the body in one direction so that the small yoga ball on the ventral side forms a continuous soothing push.The control group leaned forward on both knees (with protective gear) and supported themselves with both hands or both fists,swaying the pelvis or crawling forward.Comparing the two groups of vaginal delivery rate,maternal postural maintenance time,maternal dehvery experience,and effective correction rate of occipital position.Results The time of postoperative maintenance in the observation group was (49.51 ±10.62) minutes,and that of the control group was (18.31 ± 9.72) minutes.The difference between the two groups was statistically significant (t=6.69,P <0.05).The effective correction of abnormal fetal position rate,spontaneous delivery rate and comfort rate were 82.30% (107/130),77.69% (101/130),and 99.23% (129/130),respectively in the observation group,and 27.69% (36/130),33.85% (44/130),3.84% (5/130),respectively in the control group,the differences between the two groups were statistically significant (x2 =4.06,4.15,5.61,all P <0.05).Conclusions The undergraduate design of this sort of ball delivery group has a significant effect on the extraversion of the assisted position to the fetal position.

BACKGROUND:Modern research shows that Drynaria can delay celldegeneration and reduce the incidence of osteoarthritis. OBJECTIVE:To observe the proliferation and differentiation of rabbit bone marrow mesenchymal stem cells induced by different dosages of Drynaria freeze-dried powder, and to explore the optimum induction concentration. METHODS:Rabbit bone marrow mesenchymal stem cells were isolated and cultured in vitro by density gradient centrifugation and adherence screening methods, and then divided into blank group, positive control group (transforming growth factorβ1), high-, middle-, low-dosage Drynaria groups (0.4 mg, 0.1 mg, 5μg). Passage 3 cells were selected and cultured in different media. After 1 week, cellviability was detected by MTT method, and expression of type II col agen by immunohistochemical method. RESULTS AND CONCLUSION:Both transforming growth factorβ1 and Drynaria could improve the proliferation of bone marrow mesenchymal stem cells, and the increase in cellproliferation was ranked as fol ows:positive control group>low-dosage group>middle-dosage group>high-dosage group>blank group. Bone marrow mesenchymal stem cells were differentiated into chondrocytes under induction of transforming growth factorβ1 and Drynaria, and induced cells significantly expressed type II col agen. The expression of type II col agen was ranked as fol ows:positive control group>low-dosage group>middle-dosage group>high-dosage group>blank group. These findings suggest that Drynaria can promote the proliferation and differentiation of rabbit bone marrow mesenchymal stem cells, and the optimal dosage is 5μg.

ABSTRACT

BACKGROUND:Perioperative treatment of emergency liver transplantation for acute hepatic failure is extremely different from common liver transplantation, due to complex conditions, high risk, several complications, and high mortality. OBJECTIVE:To summarize the experience of emergency liver transplantation for acute hepatic failure during the perioperative period, and to increase the success rate in treatment of acute hepatic failure. METHODS:A retrospective analysis was undertaken on the clinical data of 38 cases undergone emergency liver transplantation for acute hepatic failure. There were 21 male and 17 female, who aged 15-69 years. Among them, 23 cases had hepatitis B virus (including 2 cases with hepatitis B and C virus), 7 cases had Wilsons disease, 3 cases had mushroom poisoning, 2 cases had unknown liver damage, 1 case had Tripterygium wilfordi poisoning, 1 case had decompensation after partial liver resection due to trauma, and 1 case had liver transplantation from corpse. RESULTS AND CONCLUSION:The survival time of the involve patients was 13-1 740 days, and the median survival time was 634 days. Perioperative survival rate was 76%, 1-year survival rate was 63%, and 2-year survival rate was 58%. During the perioperation nine cases died of brain edema and intracranial hypertension, renal failure, severe pulmonary infection, multiple organ failure, coagulation disorders (intracranial hemorrhage, upper digestive tract hemorrhage), acute respiratory distress syndrome and primary graft non-function. At present, emergency liver transplantation is stil the most effective way for acute liver failure. Hemorrhage, infection and rejection are the leading causes of the death. Each perioperative treatment is of great significance for the success of liver transplantation and long-term survival.

ABSTRACT

The present study is to investigate the protective actions of guggulsterone against the cytotoxicity produced by exposure to hydrogen peroxide (H2O2) in PC12 cells. It was evaluated by MTT [3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyl-tetrazolium bromide] reduction assay, lactate dehydrogenase (LDH) release assay, and the release of nitric oxide (NO). ROS and Ca2+ in cells were evaluated by DCFH and Fura 2-AM, respectively. Mitochondrial membrane potential (MMP) was assessed by the retention of rhodamine 123 (Rh 123). Apoptosis and morphological alteration in PC12 cells were monitored with flow cytometry and electric microscope. Vitamin E, a potent antioxidant, was employed as a comparative agent. The results showed that preincubation of PC12 cells with guggulsterone (0.1-10 μmol·L-1) prevented cytotoxicity induced by H2O2. Extracellular accumulation of LDH, NO and intracellular accumulation of ROS, Ca2+ resulting from H2O2 were significantly reduced by guggulsterone. Incubation of cells with H2O2 caused a marked decrease in MMP, which was significantly inhibited by guggulsterone. The percentage of H2O2-induced apoptosis in PC12 cells was 24.3%, and decreased in the presence of guggulsterone (0.1-10 μmol·L-1) by .8.4%, 15 .9%, 11.8%, respectively. Guggulsterone exhibited comparable potency against oxidative stress induced by H2O2 in PC12 cells as that of vitamin E. The present findings showed that guggulsterone attenuated H2O2-induced cytotoxicity, extracellular accumulation of LDH and NO, intracellular accumulation of ROS and Ca2+, loss of MMP, and apoptosis, which may represent the cellular mechanisms for its neuroprotective action.

Objective To investigate the growth inhibition of Aspergillns fumigatus by Candida albicans in vitro and to develop the selective medium for clinical isolation of Aspergillus fumigatus.Methods Aspergillus fumigatus and Candida albicans were single or co-cultured in sabouraud dextrose agar(SDA) medium and SDA broth in dark at 25 ℃,and fungal growth,pigmentation,as well as colony diameter weredocumented.Results ①The sensitivity of culture of Aspergillus fumigatus and Candida albicans on SDAplate was 100CFU/ml.②The growth of 106CFU/ml and 103CFU/ml Aspergills fumigatus was completely inhibited by 106CFU/ml Candida albicans.③Growth inhibition of Aspergillus fumigatus was correlated with the concentration of Candida albicans.④SDA containing 1 mg/L fluconazole inhibited growth of Candida albicans,and no Candida albicans was detected on SDA containing 5 mg/L and 25 mg/L fluconazole.Growth of Aspergillus fumigatus was partially inhibited on SDA containing 25 mg/L fluconazole.Conclusions Candida albicans can inhibit the growth of Aspergillus fumigatus in vitro.SDA containing 5 mg/L fluconazole can be used as the selective medium for the isolation of Aspergillus fumigatus.

ABSTRACT

Objective:To evaluate the cost-effectiveness of levofloxacin and gatifoxacin in the treatment of lower respiratory tract infections.Method:The clinical data of levofloxacin and gatifoxacin in the treatment of lower respiratory tract infections were collected with the evidence-based medicine method and were evaluated with pharmacoeconomic cost-effectiveness analysis.Result:The cure rates of two drugs were 64.29%(57.70%,70.88%),and 81.43%(70.05%, 92.81%)and the cost-effectiveness ratios were 1.90 and 5.79 respectively.The?C/?E was 20.37.Conclusion:The cost-effectiveness analysis indicated that the cost-effectivenss ratio of levofloxacin is superior to that of gatifoxacin though the cure efficacy of gatifoxacin for lower respiratory tract infections is better.

Aim To investigate the reversal effect of GP on the activity of P-gp in MDR-associated tumor cells and to develop a useful approach to inhibit the drug efflux in MDR-associated tumor cells.Methods MTT assay was used to detect the effect of GP on the cytotoxicity of cisplatin(DDP),vincristine(VCR) and doxorubicin(DOX).Flow cytometry was used to determine the influence of GP on the intracellular accumulation of DOX.Results GP synergistically increased the cytotoxity and the intracellular accumulation of DOX in K562/DOX cells;GP also increased the cytotoxity of DDP,DOX,and VCR in A549/DOX cells.However,the effect was relatively weak.Conclusion GP could increased the cytotoxity and the intracellular accumulation of chemotherapeutic drugs in MDR-associated tumor cells.GP may be a promising MDR modulator.

ABSTRACT

Objective To explore the effect of oxygen inhalation on the retinae of newborn rats and its mechanism. Methods We mimicked the retinopathy of prematurity(ROP) by putting the newborn rats in high concentrated oxygen. One-day-old rats were put into the oxygen box with the oxygen concentration of 80% for continuous 7 days; then in air condition for 7 days. The arterial blood oxygen pressure, retinal superoxide dismutase (SOD), and malondialdehyde (MDA) of the rats (1,2,4,7,8,9,11,14 days old) were examined. The diameter of retinal vessels′ main branch and the coverage rate of peripheral vessels were measured in 7- and 14-day-old rats by ink perfusion. The retinal neovascularization of rats (8,9,11,14 days old) were observed by HE staining. The rats of the same age fed in air condition were in the control group. Results The differential pressures of blood oxygen of rats (1,2,4,7 days old) in study group were significantly higher than those in the control group (P0.05). The contents of SOD of the retinae in the rats (1,2,4,7,8 days old) were significantly lower than those in the control group(P

ABSTRACT

OBJECTIVE:To evaluate the clinical efficacy,safety and status of gemcitabine for non-small-cell lung cancer (NSCLC) systematically.METHODS:Included studies retrieved from Medline,EMbase,Cancerdit,CBM and CNKI were evaluated by two reviewers alone and cross-checked.The Meta-analyses of studies were conducted using RevMan 4.2.2 software.RESULTS:12 studies containing 1,715 patients with NSCLC were included.12 studies were all reported randomly(RCT).Hidden information and application of blind method in therapy were not reported.Results of meta-analysis indicated that:(1)After chemotherapy reaction rate and the level of median disease development in GP therapy (gemcitabine combined with cisplatin) were higher than in GC therapy (gemcitabine combined with carboplatin) with similar median survival time and strong reaction of digestive tract;(2)Reaction rate;the level of median disease development and median survival time in GP therapy than in EP therapy (etoposide combined with cisplatin) but with strong toxicity in bone marrow;(3)Reaction rate of GP therapy was same to NP therapy (vinorelbine combined with cisplatin) with strong toxicity in bone marrow;(4)Reaction rate,median survival time and one-year survival rate in GP therapy were similar to in gemcitabine alone therapy with intense reaction in digestive tract;(5)Reaction rate in VGP therapy (vinorelbine,gemcitabine combined with cisplatin) was higher than in VG therapy (vinorelbine combined with gemcitabine) with strong toxicity in bone marrow but they had same the level of median disease development and median survival time;(6)Reaction rate,median survival time in DG therapy (gemcitabine combined with taxotere) were higher than in taxotere alone therapy with advanced the quality of life and strong toxicity in bone marrow;(7)Reaction rate,median survival time,the level of median disease development and one-year survival rate in therapy of gemcitabine (50 mg?min-1?30 min,i.v) were similar to that of gemcitabine (10 mg?min-1?150 min,i.v) with intense toxicity in bone marrow.17 cases of related death were reported in all researches.CONCLUSION:Gemcitabine has exact clinical efficacy and causes strong toxicity in bone marrow and reaction in digestive tract.

Objective To analyse the changes of nitric oxide and nitric oxide synthase in rat retina under acute high ocular pressure and study the effect of nitric oxide in rat retinal damage under hypertension. Methods Sixty Wistar rats were divided randomly into five groups:Ocular hypertension 30 min,60 min,90 min and 12 h,24 h after reperfusion.Elevation of the ocular pressure in the anterior chamber of the rat eye caused retina ischemic damage.The changes of retinal nitric oxide content were observed indirectly by measuring NO 2 -/NO 3 - content in retina.The distribution and changes of neuronal constitutive nitric oxide synthase (ncNOS)were studied by immunocytochemical localization of ncNOS. Results ncNOS positive neurons were distributed in the inner nuclear layer (INL),ganglion cell layer (GCL) and the inner plexiform layer of the normal and ischemic rat retina.During acute high IOP 30 min,60 min and 90 min,NO content decreased gradually and ncNOS immune activity weakens.During reperfusion,NO content increased remarkably (P

ABSTRACT

Objective To evaluate the effects and possibility of nitroglycerin on Pregnancy induced hypertension (PIH) treatment Methods In comparision with the third trimester normal pregnant women (Normal group) (n=11), 34 moderate/severe PIH cases were divided into three groups: PIH treated only with magnesium sulfate (Mg group) (n=11); PIH treated only with nitroglycerin (Ng group) (n=11); and PIH treated with Mg+ Ng (Combination group) (n=12). The endothelial nitric oxide synthase (eNOS) in placenta was investigatd immunohistochemiscally and quantitatively.
